Molecular Cell Biology: To detect the cell structures, cell skeletons, membrane fluidity, receptors, the structure and the distribution of cell organelles, apoptosis and the other cell biological inspections.
Immunology: Immunofluorescene co-localization detection, protein detection.
Clinical Services: To detect the aberrance of cells in tissue slides, tumor maker, pathogens and other biomarkers.
Plasmids construction (optional)→Cell transfection (optional)→Immunostaining→Microscopic inspection and imaging (optional to two-phonton confocal microscope)→Imaging processing.
Cell Fixation for ImmunoStaining
1. Fix cells in 4% formaldehyde (PFA) in PBS/pH 7.4 for 15 min. Alternatively, cells may be fixed in 100% methanol or in methanol/acetone mixture (1:1) at -20℃ for 6 minutes. If methanol or methanol/acetone mixture fixation is used skip to step 4.
2. Wash in PBS 3 X 10 min.
3. Permeabilize in 0.2% Triton X-100 plus 1% normal goat serum (NGS) in PBS/pH 7.3 for 5 minutes on ice.
4. Wash in PBS + 1% NGS 3 X 10 min.
5. Incubate in the appropriate concentration of primary antibody for 1 hour at room temp. in a humidified chamber. If using 22mm X 22mm square coverslips, 30 ul of diluted antibody is placed on the coverslip and the coverslip is inverted onto a glass slide. The slide is then placed in the humidified chamber which is incubated at room temp.
6. Wash in PBS + 1% NGS 3 X 10 min.
7. Incubate in secondary antibody (FITC or Texas Red conjugated) at a dilution of 1:50 for 1 hour in a humidified dark box at room temp.
8. Wash in PBS 4 X 10 min.
9. Mount cover slips with a drop of mounting medium 【Click here for Mowoil Mounting Medium】and seal coverslip with clear nail polish to prevent drying and movement under the microscope.
Customers should provide the constructs, antibodies and fluorescences dyes as indicated, otherwise provides the gene ID to Yrbio so that Yrbio can amplify the genes using the indicated vectors (customers should pay for the services of cloning and plasmid preparation).
This legend shows a investigated protein expressed for 48 hours in HeLa cells: Red for MitoDsRed indicates the mitochondria, Gene for stable expressed LC3-GFP indicates autophagosome, Blue for the immunostained the investigated protein.
The area of 2-D imaging is passed away. Biological research needs the 3D and 4D living cell imaging. Yrbio is dedicated to track a single living for cell movements or cell cycle imaging.
Optimize the Culture Enviroments of the Cell Inspection Platform
Yrbio is good at modify the cell inspection platforms so that we can controls the humidity, CO2 concentration, and the temperature as we needed during the living cell imaging.
Modify the Probes
Yrbio improves the high sensitive probes so that we take images as quickly as we can. In most circumstances, we use fluorescence protein tag bio-marker such as MitoDsRed and Tublin-GFP. Using other dyes together, Yrbio can shift the objective lens automatically and merges the videos as needed. Using the high speed cooled CCD to scan for images.
Perfect the Image Processing
Yrbio has several experts (PhD. of molecular cell biology) in imaging processing. We can program softwares and process the images with the facility of ImageJ, MatLab and other softwares.
