

The commonly structure of shRNA vector contains the GFP marker is:
pol III specific promoter(such as hU6)---shRNA---CMV promoter---EGFP---Poly A
The advantage of this kind of expression frames is universal expression and precise termination. However, pol III is a unusual and low expression polymerase, and causes nonspecific expression that disturbs the gene silencing. The mirshRNA vector is introduced to solve these problems. The structure of mirshRNA is:
pol II specific promoters (such as CMV promoter)---EGFP---mir30 left arms---shRNA---mir30 right arm---Poly A

(2)EGFP inserted before shRNA, we can use the green fluorescence intensity to detect the shRNA expression level.
(3)Using of microRNA arms can enhances efficiency of Dicer, so that promoter the silencing efficiency.
(4)The CMV promoter can be replaced other promoter pol II specific promoters, reduce the non-specific expressions.
