

Methylation Specific PCR (MSP) is a bisulfite conversion based PCR technique for the study of DNA CpG island methylation. For MSP experiment, two pairs of primers are needed (one pair specific for methylated DNA (M) and the other for unmethylated DNA (U)). To detect methylated or unmethylated DNA, in each primer (or at least one of the pair) sequence, one or more CpG sites are included. First, DNA is modified with sodium bisulfite and purified. Then, two PCR reactions are performed using M primer pair and U primer pair. Successful amplification from M pair and U pair indicated methylation and unmethylation respectively. MSP is a high sensitivily assay and is convenient to test large scale samples.
