

Yrbio shRNA constructs were designed using siRNA algorithm considering sequence, specificity and position scoring for optimal hairpins. The hairpin consists of a 21-base stem and a 6-base loop. The hairpins were cloned into the eukaryotic expression vector and sequencing verified. Yrbio possess more than 10,000 human and mouse gene, over 50,000 shRNA libraries. Experiment verified that more 90% of the shRNA are effective and the overall silencing efficiency reaches 90%. To silence the target gene Yrbio provide the replication-incompetent shRNA vector, lentiviral packing plasmids, 293T packing cells and the transfection reagents. For further researches, Yrbio provides the stable RNAi cell lines service.
The prefered pYrbio-LT-1 vector consisting the neomycin resistance gene and the GFP which can screen for stable cell line and real-time monitoring the the transfection efficiency.
A. Yrbio will design 3 strands of DNA sequences acccording to the target mRNAs, 19~21 bases in length.
B. According the target sites selected in step A, Yrbio design the sense and antisense strands with a 9nt-loop linker, which made up the short hairpin RNA.
C. NS controls.
Yrbio will introduce the BamHI and HindIII restriction enzyme cutting sites to the each side of the DNA template.
