Mowiol Mounting Medium

This is a bench top experience taken from Dr. Lu Fei (Peking University) and modified by Weiming Xiao.

Mowiol mounting media:


mowiol                 2.0 g
Glycerol                6.0 g, mix thoroughly, it's hard to dissolve
H2O                     6 ml
0.2 M Tris-Hcl (pH 8.5)  12 ml, heat to 60℃, mix until it is dissolved (It needs at least 2 h).
10 mg/ml DAPI         1 μl 
DABCO              2.5%
5000 rpm 15 min, store the supernatant at -20℃ in 1.5ml tube. Warm to use.
Mowiol: Names Mowiol 4-88,    CALBIOCHEM, Lot#B55723
DABCO: (Anti-quenching Reagents)：1，4-Diazabicyclo[2,2]octane,  SIGMA, Lot 044k3648

 

Notes: Water bath incubating during the mixing. Be patient, this is done once for all!

Usage：Immunofluorescence Mounting Medium with anti-quenching reagents (DABCO). Product M01-42-201 contains DAPI which can be used as an indicator of nuclear.
